超高效液相色谱-串联四极杆飞行时间质谱法筛查饲料中11种镇静剂类药物

Screening Method for 11 Sedatives in Feedstuff by Ultra Performance Liquid Chromatography-Quadrupole Time-of-Flight Mass Spectrometry

  • 摘要: 为了快速筛查未知饲料样品中的镇静剂类药物,应用超高效液相色谱-串联四极杆飞行时间质谱建立了筛查饲料中11种镇静剂类药物的方法。在不同添加浓度下获得了精确的分子离子质量,质量偏差的绝对值低于3×10-6。11种镇静剂类药物在0.02~1 mg/L范围内线性关系良好,相关系数r≥0.99,添加浓度为0.5~5 mg/kg时,其添加回收率在50%~120%之间,在饲料基质中的检测限为0.1 mg/kg。该方法简便、高效、准确,适于饲料中11种镇静剂类药物的分析检测需求。

     

    Abstract: A method based on ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF) was developed for quantitative analysis of 11 sedatives in feedstuff. The accurate mass was obtained in different spiking levels (from 0.5 to 5 mg/kg)and the accuracy error was lower than 3×10-6, which was well within the accepted limits for target confirmation. The linearity of response ranges from 0.02 to 1.0 mg/L, and the correlation coefficient is greater than 0.99. The average recoveries range from 50% to 120% of the fortified 11 sedatives in feedstuff at 0.5—5 mg/kg levels. and the limits of detection are 0.1 mg/kg. The results indicate that the method is easy, fasty, and sensitive. This method can meet the requirements for simultaneous determination of 11 sedatives in feedstuff.
