采用UPLC-Q-TOF MS技术分析坛紫菜中脂质成分

Rapid Identification of Lipid Compositions from Porphyra haitanensis by UPLC-Q-TOF MS

  • 摘要: 采用超高效液相色谱-飞行时间质谱(UPLC-Q-TOF MS)联用技术快速鉴定坛紫菜中脂质成分。以乙腈、甲醇、异丙醇和水作为梯度洗脱剂,Waters Acquity UPLC TM BEH C8色谱柱为固定相,快速分离坛紫菜中的脂质成分;同时,结合脂质成分的高分辨质谱裂解规律,鉴定了脂质结构,并确定了脂质的酰基位置和脂肪链长度及其不饱和度。结果表明,坛紫菜中脂质成分大致分为糖脂、磷脂和甘油三酯,其中糖脂类包括5种,分别为DGDG、Lyso-DGDG、MGDG、SQDG和Lyso-SQDG;磷脂类包括3种,分别为PG、Lyso-PG和Lyso-PC。该方法在鉴定坛紫菜的脂质成分种类和定性分析脂质成分方面具有很好的应用价值。

     

    Abstract: The lipid compositions of Porphyra haitanensis were rapidly identified by ultra performance liquid chromatography-time of flight-mass spectrometry (UPLC-Q-TOF-MS). With acetonitrile, methanol, isopropyl alcohol and water as gradient eluents, the total lipids extracted from Porphyra haitanensis were separated by Waters Acquity UPLCTM BEH C8 column. In addition, based on tandem mass spectra, the position of acyl chain, length and unsaturation of lipid chain were identified, which could be useful for lipid composition characterization. The results show that Porphyra haitanensis contains glycolipids (DGDG, Lyso-DGDG, MGDG, SQDG and Lyso-SQDG), phospholipids (PG, Lyso-PG and Lyso-PC) and triglycerides. The method has well application in qualitative and quantitative analyses of lipids from Porphyra haitanensis.
