兔血中5β-H-17-OH雄甾烷的气相色谱-质谱测定

摘要: 建立了一种气相色谱 -质谱 (GC/MS)技术测定兔血中甲睾 (MT)的主要代谢物 5 β-H-1 7-OH雄甾烷(5β-MT)质量浓度的方法 ,为药物动力学等研究提供技术方法和数据。 6只兔口服 2 0 mg MT后按设定时间点取血 ,用甲醇提取血浆 1 m L,提取液用 β-葡萄糖醛酸苷酶于 5 5℃水解 3 h。去氢甲睾 (DHMT)为内标物进行定量计算。结果显示 :给药后每个血样中均含有 5 β-MT,计算每个时间点 6只兔的 5 β-MT平均质量浓度 ρ,发现在 3 h时达到峰值 2 .2 mg/LAbstract: The determination of the essential metabolite 5β-H-17-OH-Androstane (5β-MT) from methyl testosterone (MT) in rabbit blood by gas chromatography-mass spectrometry was performed to supply method and data for pharmacokinetics study. 2mL blood taken from six rabbits ear was collected at setting time point before and after P.O. administration 20mg MT,1mL plasma sample was extracted with methanol, then the extractant was enzymed by β-glucuronidase. Quantitative analysis was performed with internal standard DHMT. The result showed that 5β-MT was presented in each blood sample, the maximal average mass concentration of six rabbit is 2.2mg/L at 3h.
