气相色谱-质谱选择离子检测法测定Melatonin的血药浓度

摘要: 本文应用GC/MS仪 ,采用内标咖啡因定量 ,选择单离子检测建立了测定Melatonin的方法 ,Melatonin的标准曲线为Y =0 .0 5 82 +0 .6 0 6 7X(r =0 .9994 ) ,其10、2 0、5 0、2 0 0、4 0 0ng/mL五浓度的血样回收率分别为 74 .4 3、93.39、94 .39、81.98、86 .10 % ;日内精密度为 6 .5 5 % ,日间精密度为 14 .5 0 %。此方法可满足测定要求 ,可用于Melatonin的药代动力学研究。应用此法测定了家兔在灌服 30mg/KgMelatonin后的血药浓度。

Abstract: A method by GC/MS-quantitative technique with coffeine as ISTD which can determine Melatonin has been developed.The standard curve of Melatonin in blood is Y=0.0582+0.6067X(r=0.9994),the recoveries of five concentrations of 10,20,50,200,400 ng/mL are 74.34,90.39,94.39,81.98,86.10% respectively;and the precision within a day is 6.55%,the precision between days is 14.50%.We determined the blood concentration of Melatonin in rabbits after 30 mg/Kg Melatonin had been administered.The method is proved to be good enough to study pharmacokinetics of Melatonin.
