Abstract:
PDCD5 protein is an important regulator in both apoptotic and non-apoptotic programmed cell death. Since the human PDCD5 protein has been shown to localize mainly in the nucleus, it is unknown whether PDCD5 protein itself directly bind DNA. A series of DNA sequence are designed for studying the interaction with PDCD5 protein. Here we show that PDCD5 protein directly binds to double-stranded DNA and singlestranded DNA by employing MALDI-TOF-MS. For the first time we observe that PDCD5 protein binds to DNA with significant preference to TC-rich sequence in gas phase. On the other side, there can be different binding sites for PDCD5 to bind with DNA.
