超高效液相色谱-电喷雾串联质谱法快速分析烟草中7种潜香物质

Rapid Analysis of 7 Aroma Precursors in Tobacco by UPLC-ESI-MS/MS

  • 摘要: 使用超高效液相色谱电喷雾串联四极杆质谱(UPLC-ESI-MS/MS)在多反应离子监测(MRM)模式下分离鉴定了烟草中7种潜香物质。烟样经甲醇提取,XAD-2柱分离纯化,选择中性丢失162 u扫描确定母离子。通过对母离子二级质谱扫描(MS2),并根据 m/z 192.7、179.2、163.1、134.7、133.2等特征碎片离子峰分析结果确定了7种潜香型物质,分别为熊果苷、香豆酸-葡萄糖苷、香豆素奎尼酸、新绿原酸、绿原酸、葡萄糖基阿魏酸、3-氧化紫罗兰醇葡萄糖苷。

     

    Abstract: A method for identification of 7 aroma precursors in tobacco was established by using ultra performance liquid chromatography-tandem mass spectrometry interfaced with electro-spray ionization(UPLC-ESI-MS/MS) in the multiple reaction monitoring(MRM) mode. Aroma precursors in tobacco were extracted with methanol, cleaned up with XAD-2 column, then detected and identified by ESI-MS/MS with the neutral loss method. According to the characteristic product ions of m/z 192.7, 179.2, 163.1, 134.7, 133.2 acquired by product ion scan(MS-2), 7 potential flavor compounds are identified, they are arbutin, trans-p-coumaric acid-4-o-β-D-glucopyrannoside, coumaroylquinic acid, neochlorogenic acid,chlorogenic acid, feruloylglycoside, 3-oxo-α-ionol-β-D-glycoside. The method is rapid, convenient and suitable for the determination of aroma precursors in tobacco leaves.
