液相色谱—质谱法(LC-MS)分析细胞色素C酶解肽谱及一级结构

Peptide Mapping and Primary Structural Analysis of Cytochrome C by Liquid Chromatography-Mass Spectrometry

  • 摘要: 本文采用液相色谱—质谱联用法(LC/MS),对一种蛋白质—细胞色素C(Cyt.C)的胰蛋白酶酶解产物进行了质量肽谱分析,确定了各肽段在蛋白质中的位置。并用串联质谱(MS/MS)分析了大部分肽段的序列,均获得满意的结果。

     

    Abstract: Liquid chromagraphy-mass spectrometry (LC/MS)was used to analyze the peptide mapping of Cyt. C digested by TPCK-trypsin for determination of the molecular weight of each peptide fragment. Most of the peptide fragments were sequenced by tandem MS (MS/MS)to obtain the amino acid sequence. The whole method of liquid chromagraphy -mass spectrometry(LC/MS)has been founded and applied to biological molecules.
