Application of Neutral Loss Scan Mode in Mass Spectrometry for Identification of the Phase Ⅱ Metabolite of Nitazoxanide in Dog Plasma

摘要: 硝唑尼特经狗口服后在体内迅速代谢成替唑尼特,随后替唑尼特进一步与葡萄糖醛酸结合生成了葡萄糖醛酸替唑尼特结合物。本实验主要运用了三重四极杆串联质谱的中性丢失扫描功能,并结合质谱的全扫描功能和酶解试验,成功地对该硝唑尼特Ⅱ相代谢物进行了鉴定。Abstract: Nitazoxanide, a new broad-spectrum parasiticidal agent was rapidly deacetylated to tizoxanide, followed by the formation of glucurono-conjugate after dog oral administration. In this experiment, neutral loss scan and full scan mode of triple quatrupole mass spectrometer and enzyme hydrolysis were used to identify the glucuronide of tizoxanide. The results show that neutral loss scan is a useful tool for identification of conjugate metabolites.
