α-腈基阿魏酸及其二元混合基体在基体辅助激光解吸/电离飞行时间质谱法测定蛋白质中的应用

  • 摘要: 本文应用自行设计、制备的新基体α-腈基阿魏酸及α-腈基阿魏酸与3-硝基苄醇组成的固液相二元混合基体,用基体辅助激光解吸/电离飞行时间质谱法成功地对蛋白质样品胰岛素,细胞色素C和白蛋白进行了分子量测定。研究结果表明,α-腈基阿魏酸可以有效地解吸分子量较小的胰岛素,而α-腈基阿魏酸与3-硝基苄醇组成的二元混合基体则能够明显地提高分析灵敏度和重现性,测得分子量较大的细胞色素C和白蛋白的激光质谱图

     

    Abstract: CyanoFerulic Acid and Its Binary Mixture as Matrices for MatrixAssisted Laser Desorption/IonizationTimeofFlight Mass Spectrometry of ProteinsAuthor Deng Huimin,Lai Zhihui,Li Jun,Zhao Shankai\=(Instrumentation Analysis & Research Centre,Zhongshan University,\=Guangzhou 510275,China)\=\=Received 1998-11-02\=\= Abstract \=\;cyanoferulic acid(CFA)designed and prepared in our laboratory and the solidliquid phase comatrix composed of cyanoferulic acid and 3nitrobenzyl alcohol(CFA+NBA) have been successfully tested for matrixassisted laser desorption/ionization timeofflight mass spectrometry of proteins.The results demonstrate that CFA can effectively desorb insulin which has lower molecular weight,and the binary comatrix CFA+NBA obviously enhances the sensitivity and reproducibility,performing MALDI spectra of cytochrome C and albumin which have higher molecular weight.\=\=Key Words: cyanoferulic acid,3nitrobenzyl alcohol,solidliquid phase comatrix,matrixassisted laser desorption/ionization,protein,molecular weight determination
