有机质谱分析在芒果苷原料药微量杂质结构鉴定中的应用
Applications of Organic Mass Spectrometry in Structure Identification of an Impurity Compound in Prepared Mangiferin Extracted from Mangifera indica L. leaves

摘要: 采用制备色谱对芒果苷原料药中的一种主要杂质进行分离纯化,利用HR-ESI(-)-MS/MS分析其结构。在芒果苷及其杂质的一级质谱中,其M-H-峰分别为 m/z 421.077 5和435.097 3;在HR-ESI(-)-MS/MS串联质谱分析中,芒果苷的主要碎片峰有 m/z 331.043 6、301.033 5、271.023 7;而杂质的主要碎片峰有 m/z 345.065 7、315.048 6、287.018 9、272.030 8。经有机质谱分析,并经 UV、1HNMR、13CNMR进一步确认,芒果苷原料药中微量杂质的结构为2-C-β-D-glucopyranosyl-1, 6, 7-trihydroxy-3-methoxy-xanthen-9-one。根据HR-ESI(-)MS/MS分析,给出了芒果苷及其杂质主要碎片的ESIMS质谱裂解途径。Abstract: An impurity compound in prepared mangiferin extracted from Mangifera indica L. leaves was isolated by high performance liquid chromatographic (HPLC). Its structure was identified by HR-ESI (-)-MS/MS and further confirmed by NMR spectroscopic studies. Based on the spectral data, the molecular structure of the impurity is identified as 2-C-β-D-glucopyranosyl-1, 6, 7-trihydroxy-3-methoxy-xanthen-9-one. The fragmentation behaviors of mangiferin and the impurity are proposed.
