Abstract:
Identification and Quantitation of Fusarium Metabolites in Rice Cultures of Fusarium Fungi by GC/MSLuo Yi;Liu Feng; Feng Jianlin; Hu Xuyin; Yang Jinsheng(Institute of Pharmacology and Toxicology,Academy of Military Medical Sciences,Beijing 100850,China)Kuang Kaiyuan;Shi Shiyin(Institute of Plant Protection,Shanghai Academy of Agriculture Sciences, Shanghai 201 106,China)Abstract:In this paper,it is mainly reported on results of identification and quantitation of toxic metabolites in rice culture of Fusarium fusarioides(Frag. & Cif. )by GC/MS/El+,which was isolated from the ear of red scab wheat. Rice culture of Fusarium fusarioides(Frag. &Cif) was extracted with acetonitrile: water (3:1, V/V)followed by defatted with n-he-cane. The extract was cleaned up on the column of Florisil and eluted with chloroform: methane(9:1,V/V). The eluate was concentrated to dryness underreduced pressure. The residue was derivatizrf with heptafluorobutyrylimidazole(HFBIM) followed by GC/MS analysis. By the identification of GC/MS,it was found that in the culture extract Fusarium fusarioides(Frag. & Cif. ) produced the secondary toxic metabolites, such as T-2 tetraol, T-2 triol, 3'-hydroxyl T-2 toxin, neosolaniol(NS),HT-2 toxin and T-2 toxin etc. The contents of these produced toxins were quantitatively determined with selected single ion, respectively,compared with standard of each toxin.It was confirmed that the strain of Fusarium fusarioides(Frag. & Cif. )mainly produced those trichothecenes belonged to T-2 toxin group. The results are of significant for the study on Fusarium fungi and their toxic metabolites.Keywords: Fusarium fusarioides (Frag. & Cif. ), trichothecenes, GC/MS,culture
